Description

A DNA profile, also called DNA typing, DNA fingerprinting or genotyping, is a genetic record unique to an individual horse. It is built from a set of genetic markers that each horse inherits half from its sire and half from its dam. A profile does not identify breed. It identifies the individual horse, and it can be compared with the profiles of other horses to confirm or rule out a parent–offspring relationship.

DNA profiles are used for breed registration, for parentage and relationship testing, and as a permanent DNA record that can positively identify a horse if it is ever lost or stolen. Animal Genetics uses pulled mane or tail hair as the sample.

Our marker panel

Our equine profile uses a multiplex panel of 20 markers, following the standards of the International Society for Animal Genetics (ISAG) and leading laboratories including the University of California, Davis and the University of Kentucky. The panel includes the ISAG core microsatellite (STR) markers, additional industry-standard STR markers, and the AME marker, which confirms the horse’s sex:

AHT4, AHT5, ASB2, ASB17, ASB23, CA425, HMS1, HMS2, HMS3, HMS6, HMS7, HTG4, HTG6, HTG7, HTG10, LEX3, LEX33, UM011, VHL20 and AME.

Because we report results using the international alphanumeric nomenclature, our equine profiles can be compared directly with profiles produced by other laboratories using the same markers.

Parentage testing

For parentage testing, the foal’s profile is compared marker by marker with the profiles of its proposed sire and dam. A foal must share one allele with each true parent at every marker, so any mismatch shows that a proposed parent is excluded. Our panel has a combined exclusion probability greater than 99.99% for detecting incorrectly assigned parentage.

Testing the foal and the proposed sire alone can verify the sire. Testing the foal with both parents gives the most reliable result, because the dam’s profile accounts for the half of the foal’s DNA that did not come from the sire.

Our crosschecking procedures for sample processing and recording ensure a high level of accuracy and quality control, and all parentage conclusions are provided in writing once testing is complete.
